Triptorelin description 

Deca-peptide and a gonadotropin-releasing hormone agonist (GnRH agonist) Triptorelin acts as an agonist analog of Gonadotropin-releasing hormone, thus reversibly repressing of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Triptorelin is structurally derivative of the gonadotropin-releasing hormone. The distinguishing feature between the medicinal substance and the naturally occurring hormone is the amino acid in position 6. In GnRH, glycine is in this position, whereas in triptorelin there is tryptophan. 

How does Triptorelin work?

Triptorelin mimics the action of natural GnRH: Triptorelin binds to receptors in the pituitary gland and stimulates the secretion of gonadotropins (specifically luteinizing hormone and follicle stimulating hormone). This initially causes an initial phase of very strong stimulation of LH and FSH. But further use is followed by strong down-regulation of gonadotropin-releasing hormone receptors, thereby reducing long-term gonadotropin release, leading to inhibition of androgen and estrogen production. The result can be a reduction of serum testosterone to levels which are typically seen in surgically castrated men. Ultimately, tissues and functions that require these hormones become inactive. The effects of triptorelin can usually be reversed once Triptorelin is discontinued.

Triptorelin medical use

Triptorelin can be used to treat prostate cancer (PCa) as part of androgen deprivation therapy (ADT). Another ADT injectable medicine used to treat PCa includes Lupron which also targets the pituitary. Triptorelin is commercially available as an injection preparation. 

Triptorelin to restore testosterone production and PCT

Some people, especially bodybuilders, also discuss the very strong ability of triptorelin to restore testosterone production in cases of hypogonadotrophic hypogonadism, such as caused by high doses or long-term use of anabolic steroids (use of Triptorelin for so-called PCT, post-cycle therapy after steroid use). Several steroid users on the Internet reported that, thanks to the proper use of Triptorelin, they were able to resume their lost testosterone production even in severe and long-lasting cases of reduced or completely suspended testosterone production. If the use of triptorelin to restore testosterone production is considered, 100 μg of triptorelin should be administered subcutaneously or intravenously in a single dose and only once (because if the first dose has a very strong ability to kick testosterone again, subsequent doses or long-term triptorelin use should be led to reduced regulation of LH and FSH levels and extreme suppression of testosterone production).

Triptorelin dosage

Usual dose of Triptorelin for medical purposes in the treatment of prostate cancer (to reduce regulation of LH and FSH levels, lower serum testosterone and suppress testicular steroid biosynthesis): An injection of 3.75 mg triptorelin is usually given every 4 weeks, 11.25 mg triptorelin every 12 weeks. weeks or an injection of 22.5 mg triptorelin is usually given every 24 weeks.

Bodybuilders and steroid users who consider use triptorelin to restore testosterone production (for PCT), recommend that 100 μg of triptorelin should be administered subcutaneously or intravenously in a single dose and only once.

Warning: Overdose, abuse or improper use of Triptorelin leads to a decrease in the level of LH and FSH and subsequent hypogonadism – reduced functional activity of the gonads, which leads to a reduction in the production of estrogen and testosterone.
